Christos Kyriakopoulos, MD, University of Wisconsin, Madison, WI, discusses the prognostic and predictive impact of circulating tumor cells (CTC) and AR-V7 in the CHAARTED2 trial (NCT03419234) of abiraterone with or without cabazitaxel in metastatic castration-resistant prostate cancer (mCRPC) post-docetaxel. Baseline detection of CTC and AR-V7 correlated with poorer progression-free survival. Conversion to negative status at cycle 5, day 1 was associated with better outcomes. While AR-V7 positivity indicated potential benefit from combination therapy, statistical significance was not reached.
