ESMO 2026 | Novel AR-targeted therapies in prostate cancer: CYP11A1 inhibitors, AR degraders and RIPTACs

Robert Jones, MD, PhD, University of Glasgow, Glasgow, UK, discusses novel androgen receptor (AR)-targeted therapies for advanced prostate cancer, including steroid hormone biosynthesis inhibitors like opevesostat, AR degraders, and regulated induced proximity targeting chimeras (RIPTACs), a new class of drugs which bind to the AR and BRD4 to induce cell killing. Dr Jones highlights the potential of these therapies to overcome resistance to current AR-targeted treatments and notes that early data for the RIPTAC JNJ-6143 shows high response rates and good tolerability. This interview took place at the European Society for Medical Oncology (ESMO) 2026 Congress in Madrid, Spain.
