Silke Gillessen, MD, Oncological Institute of Southern Switzerland, Bellinzona, Switzerland, discusses findings from the Phase III STAMPEDE trial (NCT00268476), which evaluated whether adding metformin to standard of care improves outcomes in non-diabetic patients with metastatic hormone-sensitive prostate cancer. While the addition of metformin did not significantly extend overall survival, it was associated with a reduction in adverse metabolic effects linked to androgen deprivation therapy. The safety profile was consistent with known effects of metformin, primarily involving gastrointestinal toxicity, and supports further consideration of metabolic management strategies in this setting.
