ESMO GI 2026 | TRIPP-FFX: targeted intratumoral P-32 microparticles with FOLFIRINOX in uLAPC

Michele Milella, MD, University of Verona, Verona, Italy, discuss the trial design and results from the randomized non-comparative TRIPP-FFX study (NCT05466799) evaluating the feasibility and tolerability of induction FOLFIRINOX chemotherapy with targeted intratumoral placement of phosphorus-32 (P-32) microparticles in unresectable locally advanced pancreatic cancer (uLAPC). Patients were also randomized to FOLFIRINOX chemotherapy alone to provide a calibration but not comparative arm. The local disease control rate (DCR) at 16 weeks was 82% in the intent-to-treat population from the combination arm and 56% in the per-protocol population. No new toxicity signals were observed and 6 adverse events were reported that were related to implantation. In the per-protocol population, the progression-free survival, overall survival and time to local disease progression were numerically higher in the combination arm than the FOLFIRINOX alone arm, however as this was a non-comparative study, the significance of this was not assessed. This interview took place at the European Society for Medical Oncology (ESMO) Gastrointestinal Cancer 2026 Congress in Munich, Germany.
