VJOncoAlert Journal Club | PACE: ctDNA landscape after CDK4/6 inhibitor progression for metastatic HR+/HER2- breast cancer

In this latest episode of the VJOncoAlert Journal Club, Rinath Jeselsohn, MD, Dana-Farber Cancer Institute, Boston, MD, discusses findings from the PACE trial (NCT03147287) analyzing circulating tumor DNA (ctDNA) dynamics in patients with HR+/HER2- metastatic breast cancer progressing on endocrine therapy plus CDK4/6 inhibition. Frequent alterations included ESR1, TP53, and PIK3CA mutations, with several variants linked to shorter progression-free survival. Longitudinal profiling showed enrichment of resistance-associated mutations at progression, highlighting the potential of ctDNA biomarkers to guide post-CDK4/6 inhibitor treatment strategies.
