Trastuzumab deruxtecan or ramucirumab plus paclitaxel in gastric cancer
In this latest episode of the VJOncoAlert Journal Club, Kohei Shitara, MD, National Cancer Center Hospital East, Kashiwa, Japan, discusses findings from the DESTINY-Gastric04 study (NCT04704934). The study compared trastuzumab deruxtecan (T-DXd) to the combination of ramucirumab plus paclitaxel as second-line treatment for patients with HER2-positive unresectable or metastatic gastric cancer or gastroesophageal junction adenocarcinoma. Results showed that T-DXd provided superior overall survival compared to the ramucirumab-paclitaxel combination, while also delivering improved progression-free survival and higher response rates. The safety profile of T-DXd was consistent with earlier studies, although patients experienced higher rates of interstitial lung disease and pneumonitis compared to the control group.
