ASCO 2024 | Safety and efficacy of RP2 alone or with nivolumab for uveal melanoma

Joseph Sacco, PhD, MBChB, The Clatterbridge Cancer Centre NHS Foundation Trust, Birkenhead, UK, shares data on RP2, an enhanced-potency oncolytic HSV-1 therapy. The Phase II study (NCT04336241) evaluated RP2 monotherapy and RP2 combined with nivolumab in 17 patients with metastatic uveal melanoma (MUM). The objective response rate was 29.4%, with durable partial responses in 5 patients. Treatment was well-tolerated, with no grade 4/5 adverse events. Biomarker analysis showed increased PD-L1 expression and CD8+ T-cell infiltration in tumors, along with T-cell receptor repertoire changes. These findings suggest that RP2 alone or with nivolumab is a promising treatment for MUM, warranting further clinical trials. This interview took place during the 2024 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
