Giovanni Randon, MD, Fondazione IRCCS Instituto Nazionale dei Tumori di Milano, Milan, Italy, reflects on the ARMANI trial (NCT02934464), which investigated ramucirumab plus paclitaxel or CAPOX/FOLFOX followed by fluoropyrimidine monotherapy as switch maintenance or early second-line therapies in HER2-negative (HER2-) gastric/gastroesophageal junction (G/GEJ) cancer. Whilst the rate of grade 3 adverse effects (AE) doubled in the ramucirumab arm, the rate of neurotoxicity was not increased. Overall, there was a consistent safety profile of paclitaxel plus ramucirumab, making it suitable to progress into further studies.
