ESMO Asia 2024 | Latest advances in biliary tract cancer: insights from the TOURMALINE and HERIZON-BTC-01 trials
Lorenza Rimassa, MD, IRCCS Humanitas Research Hospital, Milan, Italy, discusses the Phase IIIB TOURMALINE (NCT01564537) study and the Phase IIb HERIZON-BTC-01 (NCT04466891) trial, both focusing on biliary tract cancer (BTC). The results of TOURMALINE demonstrate the potential benefits of combining durvalumab with gemcitabine-based regimens and the safety of reducing durvalumab infusion time demonstrated, which was shown to be similar to the original 60-minute infusion time. The HERIZON-BTC-01 trial’s updated analyses and overall survival for Cohort 1 (HER2-amplified with either immunohistochemistry 2+ or 3+) demonstrate extended response with an encouraging median overall survival for patients from Asia with previously treated HER2-positive BTC. These results provide reassurance for the ongoing Phase III HERIZON-BTC-302 (NCT06282575) trial investigating zanidatamab in combination with standard-of-care therapy in the first-line setting for patients with HER2-positive BTC.
