GU Cancers 2025 | STAMPEDE: updates in evaluating tE2 patches with ARPIs in prostate cancer

Nicholas James, MBBS, PhD, FRCP, FRCR, Institute of Cancer Research, London, UK, explores findings from the Phase II STAMPEDE trial (NCT00268476) of transdermal estradiol (tE2) patches versus luteinising hormone releasing hormone analogues (LHRHa) when combined with androgen receptor pathway inhibitors (ARPIs) in metastatic prostate cancer. PSA responses were similar between groups, supporting tE2 as a viable alternative for androgen deprivation therapy. Notably, tE2 was associated with fewer hot flushes and lower hypertension rates, providing an alternative toxicity profile that may improve patient quality of life. This interview took place at the ASCO GU Cancers Symposium 2025 in San Francisco, CA.
