ELCC 2025 | FAME: metformin and fasting-mimicking diet in LKB1-inactive NSCLC

Mario Occhipinti, MD, PhD, Fondazione IRCCS – Istituto Nazionale dei Tumori, Milan, Italy, discusses the Phase II FAME trial (NCT03709147), which assessed metformin with or without a fasting-mimicking diet alongside first-line chemo-immunotherapy in advanced LKB1-inactive non-small cell lung cancer (NSCLC). Despite early closure due to low recruitment, results suggest a potential survival benefit in patients with Stk11 mutations. Findings highlight the role of metabolic interventions in addressing treatment-resistant NSCLC, warranting further investigation into glucose deprivation strategies to enhance therapeutic efficacy. This interview took place at the European Lung Cancer Congress (ELCC) 2025 in Paris, France.
