ASCO 2025 | High-risk cutaneous SCC trials at ACSO 2025: KEYNOTE-630 & C-POST

Kevin Harrington, MD, PhD, FRCP, FRCR, FRSB, The Institute of Cancer Research and The Royal Marsden NHS Foundation Trust, London, UK, discusses outcomes from two Phase III trials evaluating adjuvant PD-1 inhibition in high-risk cutaneous squamous cell carcinoma (CSCC). The Phase III C-POST trial (NCT03969004) showed that cemiplimab significantly reduced recurrence risk across both nodal and non-nodal high-risk subgroups, marking the first positive trial of adjuvant systemic therapy in this setting. In contrast, the Phase III KEYNOTE-630 trial (NCT03833167) found that pembrolizumab did not significantly improve recurrence-free survival compared with placebo. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
