ASCO 2025 | DATO-Base: investigating Dato-DXd in HER2- breast cancer brain metastases

Paolo Tarantino, MD, Dana-Farber Cancer Institute, Boston, MA, provides an overview of the Phase II DATO-Base trial (NCT06176261) of datopotamab deruxtecan (Dato-DXd) in patients with HER2-negative metastatic breast cancer and active brain metastases or leptomeningeal disease. This open-label, multicenter trial includes three cohorts based on hormone receptor status and disease location. Primary endpoints focus on intracranial response, with translational analyses of blood and cerebrospinal fluid. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
