ESMO Asia 2025 | Challenges and strategies for off-the-shelf allogenic CAR T-cell therapies

Han Chong Toh, MD, National Cancer Centre Singapore, Singapore, comments on the challenges associated with off-the-shelf allogenic T-cell therapies, particularly the host versus graft response, where the body’s immune system fights back against foreign cells. Strategies to manipulate these cells to make them less detectable by the body’s immunity include the use of NK CAR T-cells, which may have fewer side effects. They are being engineered to down-regulate detectable molecules, essentially operating in a “stealth mode”. This interview took place at 2025 European Society for Medical Oncology (ESMO) Asia Congress in Singapore, Singapore.
