ASCO 2026 | Development of a CHAI–powered predictive biomarker for 1L chemotherapy intensification in mCRC

Paolo Ciraci, MD, University of Pisa, Pisa, Italy, discusses the development and validation of a computational histology artificial intelligence (CHAI)–powered predictive biomarker for first-line chemotherapy intensification in metastatic colorectal cancer (mCRC). The biomarker, validated in prospective randomized Phase III trials, can predict the benefit of intensifying upfront treatment with triplet therapy versus doublet therapy, resulting in approximately six months of advantage in median progression-free survival (PFS) and 10 months of improvement in overall survival (OS) for biomarker-positive patients. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
