Michel Ducreux, MD, PhD, Paris-Saclay University, Paris, France, discusses pharmacokinetic and biomarker analyses from the Phase II GLEAM study (NCT03816163) of zolbetuximab plus gemcitabine and nab-paclitaxel in claudin 18.2-positive metastatic pancreatic adenocarcinoma (PAC). Drug exposure was comparable to that seen in gastric and gastroesophageal junction cancer, suggesting reduced efficacy in pancreatic cancer relates to disease- or chemotherapy-specific factors. Zolbetuximab increased immune-related cytokine levels during treatment, with greater interleukin-18 elevation associated with longer overall survival.
