ESMO GI 2026 | Futibatinib, zimberelimab, and chemotherapy in first-line biliary tract cancer

Lorenza Rimassa, MD, Hunimed University, Milan, Italy, discusses a Phase I study (NCT04999761) of futibatinib, an FGFR1-4 inhibitor, with zimberelimab, an anti-PD-1 antibody, and chemotherapy as first-line treatment for unresectable advanced or metastatic biliary tract cancer, regardless of FGFR status. The combination showed a manageable safety profile with expected hematologic toxicities and encouraging antitumor activity, including a high response and disease control rate. These results support a planned Phase III trial comparing this regimen with investigator-choice checkpoint inhibitor plus chemotherapy. This interview took place at the European Society for Medical Oncology (ESMO) Gastrointestinal Cancer 2026 Congress in Munich, Germany.
