Eva Ciruelos, MD, PhD, Hospital 12 de Octubre, Madrid, Spain, shares insights from the Phase II DESTINY-Breast12 trial (NCT04739761), involving 500 patients with HER2-positive, hormone receptor-positive (HR+/HER2+) breast cancer. Notably, 50% of patients in the trial had brain metastases, either stable or active. In patients with brain metastases, trastuzumab deruxtecan (T-DXd) achieved an overall response rate (ORR) and median progression-free survival (PFS) similar to those without brain metastases. This reinforces T-DXd as the preferred second-line treatment for HER2+ breast cancer, regardless of brain metastasis status.
