ESMO Breast 2025 | Phase II trial of T-DM1 and palbociclib in metastatic HER2+ breast cancer

Pavani Chalasani, MD, MPH, The George Washington University, Washington, DC, discusses results from a Phase II study (NCT03530696) of trastuzumab emtansine (T-DM1) with palbociclib in metastatic HER2+ breast cancer. The trial, initially randomized, was modified to a single-arm design due to low accrual. The combination showed longer progression-free survival and higher response rates compared to historical data for T-DM1 alone, with consistent outcomes across estrogen receptor subtypes. No new safety concerns emerged, and the findings support further investigation in a larger randomized setting. This interview took place at the 2025 European Society for Medical Oncology (ESMO) Breast Cancer Annual Congress in Munich, Germany.
