Patrizia Giannatempo, MD, Fondazione IRCCS Istituto Nazionale dei Tumori, Milan, Italy, discusses the ongoing ARCADIA Phase II trial (NCT03824691) evaluating the combination of durvalumab and cabozantinib in patients with variant histology tumors who have received prior treatments. The trial includes a significant number of patients with neuroendocrine and adenocarcinoma subtypes. Early findings indicate a median overall survival of approximately 12 months and a median progression-free survival of 6 months, with an objective response rate (ORR) of around 42%. Notably, the ORR in patients with variant histology may exceed 40%, highlighting the potential of this combination therapy in challenging cases.
