The Phase I DAD trial (NCT04724018) initially demonstrated promising response rates in patients with treatment-naïve metastatic urothelial carcinoma receiving sacituzumab govitecan with enfortumab vedotin. Bradley McGregor, MD, Dana-Farber Cancer Institute, Boston, MA, comments on the DAD-IO trial, an extension of the aforementioned trial, which will assess sacituzumab govitecan, enfortumab vedotin, and pembrolizumab in the first line setting. Due to toxicity concerns, doses of sacituzumab govitecan will be lower than that given in the TROPiCS-04 trial (NCT04527991).
