ASCO 2022 | The mechanism of action of TPST-1120, a PPARα inhibitor

Fatty acid oxidation is often upregulated in tumors, as it promotes tumor proliferation, angiogenesis and an immunosuppressive tumor microenvironment. Mark Yarchoan, MD, Johns Hopkins Medicine, Baltimore, MD, gives an overview of the mechanism of action of TPST-1120, oral therapy targeting peroxisome proliferator-activated receptor alpha (PPARα). PPARα is a transcription factor that regulates fatty acid oxidation and TPST-1120 will subsequently inhibit tumor growth and reprogram the tumor microenvironment.
