ESMO Immuno-Oncology 2025 | Multi-specific antibodies to overcome limits of cancer immunotherapy

So we always do functional immunology in a very multifactorial manner. We do multiple pathways. We often do a lot of synergistic additive analysis studies. But when it comes to designing of immunotherapy, in the past, at least, we always ended up with a monotherapy approach. So you did such a lot of work around the pathway from surface until the nucleus, but then you ended up with a single antibody targeting that, hoping that all of those complicated pathways just kind of come together under the umbrella of that single antibody...

But we have discovered that this might happen for some dominant targets like PD-1, PD-L1, CTLA-4, but we have also discovered that this does not always get transferred to certain others like TIGIT, LAG3, and so on. So I think in this particular sense, it’s very exciting that the field currently is moving towards multi-specific antibodies, because I believe that they will actually really unlock the kind of immunology that we have been functionally doing, but never actually translating it into the clinic. So we have been also, in fact, working on the field of multispecifics and the specialty of courses that you’re now really engaging multiple targets at the same amount of time. So you’re really exploiting the pathway that we have studied for so long, but in its entirety, not by a single component. So if there are redundancies, positive redundancies, but also negative redundancies, you’re actually now simultaneously targeting this. So this is something that I think is a very exciting development that has really taken steam in this year, also in terms of market and pharma interest. And I also plan to discuss our first-in-class tri-specific antibody that we have created, which is supposed to overcome refractory tumors in patients eventually.
