AACR 2025 | First-in-class Cbl-b inhibitor GRC 65327 in solid tumors

Mario Perro, PhD, Ichnos Glenmark Innovation, New York, NY, discusses the preclinical profile of GRC 65327, a highly selective and potent Cbl-b inhibitor targeting immune suppression in the tumor microenvironment. GRC 65327 enhances T and NK cell activation, restores exhausted T cell function, and potentiates anti-PD-1 responses in preclinical models of solid tumors. The compound demonstrated favorable safety, minimal off-target activity, and robust antitumor activity, both alone and in combination therapy.
