SABCS 2024 | Overcoming resistance to HER2-directed ADCs in HER2+ breast cancer

Jangsoon Lee, PhD, University of Hawai’i Cancer Center, Honolulu, HI, explores mechanisms of resistance to HER2-antibody-drug conjugate (ADC) therapies, such as trastuzumab emtansine (T-DM1) and trastuzumab deruxtecan (T-DXd), and potential strategies to overcome them. Synthetic lethal screening can identify DNA damage repair pathways as key therapeutic targets and combining T-DXd with elimusertib, ATR inhibitor, has demonstrated significant synergy in preclinical models. This interview took place at the 2024 San Antonio Breast Cancer Symposium (SABCS) congress in San Antonio, TX.
