Guru Sonpavde, MD, AdventHealth Cancer Institute, Orlando, FL, gives an overview of a Phase I/Ib trial (NCT06631521) evaluating neoadjuvant relugolix and darolutamide before radical prostatectomy in high-risk localized prostate cancer. This all-oral regimen aims to enhance operability, allow rapid testosterone recovery, and serve as a foundation for future combination strategies. Secondary objectives include clinical and pathological response, undetectable PSA nadir rates, and post-operative recovery.
