AACR 2025 | Characterizing TP53 Y220C mutations across solid tumors

Alison Schram, MD, Memorial Sloan Kettering Cancer Center, New York, NY, describes the clinical and genomic landscape of TP53 Y220C-mutated solid tumors. This hotspot mutation, present in a small proportion of tumors, alters protein conformation and is targetable by emerging small-molecule reactivators. The analysis highlights its prevalence across cancer types, especially in high-grade serous ovarian cancer, and shows genomic similarity to other TP53-mutant tumors. This interview took place at American Association for Cancer Research (AACR) Annual Meeting 2025 in Chicago, IL.
