ASCO 2025 | DESTINY-Gastric05: first-line T-DXd ± pembrolizumab in HER2+ gastric cancer

Kohei Shitara, MD, National Cancer Center Hospital East, Kashiwa, Japan, comments on the design of the Phase III DESTINY-Gastric05 trial (NCT06731478), comparing first-line trastuzumab deruxtecan (T-DXd) plus chemotherapy with or without pembrolizumab versus standard trastuzumab-based regimens in HER2-positive unresectable or metastatic gastric or gastroesophageal junction cancer. The study targets patients with PD-L1 CPS ≥1, aiming to assess progression-free and overall survival outcomes. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
