ESMO 2026 | HERIZON-GEA-01: zanidatamab + chemotherapy ± tislelizumab shows OS benefit in HER2+ mGEA

Kohei Shitara, MD, National Cancer Center Hospital East, Kashiwa, Japan, discusses the second interim analysis findings from the HERIZON-GEA-01 (NCT05152147) trial, which evaluated zanidatamab plus chemotherapy with or without tislelizumab compared with trastuzumab plus chemotherapy in HER2-positive (HER2+) advanced/metastatic gastroesophageal adenocarcinoma (mGEA). The analysis demonstrated significant overall survival (OS) improvement with both the zanidatamab doublet and triplet regimens compared with chemotherapy plus trastuzumab, with a median OS of 22.9 months and 27 months for the investigational regimens, respectively. Dr Shitara notes that while the comparison between the doublet and triplet did not reach statistical significance, the triplet regimen showed a trend towards improved OS. This interview took place at the European Society for Medical Oncology (ESMO) 2026 Congress in Madrid, Spain.
