ASCO 2025 | Assessing a DNAJ-PKAc-targeting therapeutic vaccine in FLC

Marina Baretti, MD, Johns Hopkins University School of Medicine, Baltimore, MD, discusses a Phase I (NCT04248569) study of a therapeutic vaccine targeting the DNAJ-PKAc chimera in combination with nivolumab and ipilimumab in patients with fibrolamellar hepatocellular carcinoma (FLC). The trial assessed safety and immunogenicity, with most evaluable patients developing T cell responses against the neoantigen. Disease control and partial responses were observed, particularly in those with robust immunologic activation. Findings support further development of neoantigen-directed immunotherapies in this rare cancer. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
