ASCO 2025 | ctDNA detection and outcomes in the CALLA trial for cervical cancer

Jyoti Mayadev, MD, University of California San Diego Medical Center, San Diego, CA, discusses the Phase III CALLA trial (NCT03830866), which evaluated the prognostic utility of ultrasensitive circulating tumor DNA (ctDNA) analysis in locally advanced cervical cancer. Using a tumor-informed minimal residual disease (MRD) assay, ctDNA was tracked at baseline and post-treatment in patients receiving durvalumab with chemoradiotherapy (CRT) or CRT alone. High baseline ctDNA levels and persistent detection post-treatment were associated with increased relapse risk, while reduced ctDNA detection correlated with improved survival, particularly in patients with high PD-L1 tumor expression. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
