Omid Hamid, MD, The Angeles Clinic and Research Institute, Los Angeles, CA, comments on the Phase II SWOG S2000 trial (NCT04511013) of encorafenib, binimetinib, with nivolumab versus ipilimumab and nivolumab in patients with symptomatic BRAFV600-mutant melanoma brain metastases. The triplet regimen demonstrated improved progression-free survival compared to the immunotherapy doublet, meeting the primary endpoint. Response rates were higher with the triplet approach, with toxicity profiles consistent with prior studies. These findings suggest a potential role for targeted plus immunotherapy in this challenging patient population.
