ASCO 2025 | Cervical cancer updates at ASCO 2025: OUTBACK and KEYNOTE-A18

Jyoti Mayadev, MD, University of California San Diego Medical Center, San Diego, CA, provides an overview of recent trials in cervical cancer including the Phase III OUTBACK (ACTRN12610000732088) and Phase III KEYNOTE-A18 (NCT04221945) trials. The OUTBACK trial demonstrated that patients still experience sexual heath and symptom-related problems long after treatment, and the KEYNOTE-A18 trial continues to show the benefits of pembrolizumab with chemoradiotherapy in high-risk locally advanced cervical cancer. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.

We had a really great session today on cervical cancer. So Dr Mileshkin presented her long-term results from the OUTBACK trial. And she really showed that patients continue to suffer with sexual side effects, that about 30 to 40% of patients still have difficulty with vaginal dryness, have potential concerns about pain with intercourse. And I think these patient-reported outcomes are so important because as we keep increasing the cures for cervical cancer, we’re going to have patients survive and survive longer...

And I think these patient-reported outcomes are so important because as we keep increasing the cures for cervical cancer, we’re going to have patients survive and survive longer. And we need to know how we can help them. Another trial that was presented was by Dr Duska. And she presented the long-term outcomes from KEYNOTE-A18, which is the trial that looked at chemoradiation and pembrolizumab versus chemoradiation and placebo. And at now three years, they show in their final analysis, there continues to be separation of the curves in terms of progression-free survival and overall survival, and their intent-to-treat population with locally advanced cervical cancer, showing a benefit for IO therapy, specifically for pembrolizumab, during and after chemoradiation. So those were two trials that I looked at with a focus and a lens on cervical cancer here at ASCO.
