ESMO 2025 | Novel therapies for poorly differentiated neuroendocrine carcinomas

Aman Chauhan, MD, Sylvester Comprehensive Cancer Center, Miami, FL, highlights the limited progress in new drug development for poorly differentiated neuroendocrine carcinomas, with few treatment options available, particularly for extrathoracic cases. Emerging therapies, such as DLL3-targeted T-cell engagers and oncolytic virus combinations with ipilimumab, offer potential hope for addressing this unmet need. This interview took place at the European Society for Medical Oncology (ESMO) 2025 Congress in Berlin, Germany.

We have not made any movement in terms of new drug development when it comes to poorly differentiated neuroendocrine carcinomas. There is some light at the end of the tunnel for small cell lung cancers, but extrathoracic, poorly differentiated neuroendocrine carcinomas. Unfortunately, very few treatment options exist. There have been some movements in this space with DLL3-targeted T-cell engagers being developed, and then our study is also looking to bridge those gaps...

There have been some movements in this space with DLL3-targeted T-cell engagers being developed, and then our study is also looking to bridge those gaps. Hopefully, the oncolytic virus combination with ipilimumab will help move the drug development in the right direction in this unmet need.
