AACR 2026 | Zedoresertib and lunresertib shows activity in genomically altered tumors

Timothy Yap, MBBS, PhD, FRCP, The University of Texas MD Anderson Cancer Center, Houston, TX, explores data from a Phase I trial (NCT04855656) of zedoresertib with lunresertib in advanced solid tumors harboring CCNE1, FBXW7, or PPP2R1A alterations. The first-in-class synthetic lethal combination of WEE1 and PKMYT1 inhibitors demonstrated a manageable safety profile and encouraging early antitumor activity. Pharmacokinetic and pharmacodynamic analyses confirmed target engagement, while circulating tumor DNA responses support continued dose optimization and further clinical development. This interview took place at the American Association for Cancer Research (AACR) Annual Meeting 2026 in San Diego, CA.
