Timothy Yap, MBBS, PhD, FRCP, The University of Texas MD Anderson Cancer Center, Houston, TX, discusses the background for a Phase I/II study (NCT07300943) of CLIO-8221, a HER2-targeted, dual-payload antibody-drug conjugate (ADC) conjugated to both topoisomerase 1 inhibitor (TOPi) and ATR inhibitor (ATRi), in patients with advanced HER2-expressing solid tumors. In preclinical models, CLIO-8221 has demonstrated potent anti-tumor activity and a good safety profile and is being evaluated as a monotherapy in the dose-escalation Phase I portion of the study across HER2-expressing solid tumors. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
