ASCO 2026 | RASolute 302: daraxonrasib in metastatic pancreatic ductal adenocarcinoma

Zev Wainberg, MD, University of California, Los Angeles, CA, comments on the significance of the RASolute 302 (NCT06625320), which demonstrated the effectiveness of daraxonrasib in improving survival in metastatic, pre-treated pancreatic ductal adenocarcinoma (PDAC). The trial showed a consistent result across the board, with daraxonrasib outperforming chemotherapy in terms of response rate, progression-free survival, and overall survival, marking a transformational moment in the space. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.

Sure, yes, happy to talk about it, having been involved with it for some time. It’s been great to see it through the finish line. Really a transformational moment, in my opinion, in the history of pancreatic cancer, because for the first time, we’re able to show that something other than chemotherapy has effectiveness at improving survival, which is no small feat, as you know. So, you know, we’ve known for a few years that this drug has some activity. Early in the phase one study, we saw it very early on, reported as such. But the randomized phase three trial, the RASolute 302 trial, was the ultimate test and splitting the patients 50-50 into chemotherapy or the pill was the first logical trial to do with this drug. And after patients received chemo, we know that the chemo only gets you so far and obviously the study was powered for a survival advantage. That’s exactly what the RASolute 302 study showed. The group of patients who received daraxonrasib outperformed the group of patients who received chemotherapy in every aspect, response rate, progression-free survival, more importantly, overall survival, doubling the overall survival from about 6.7 to 13.2. So really a consistent result across the board. It also made those patients feel better because even despite side effects, which are notable, the quality of life improvement scale was significantly better in the group of patients who received eribulin over those who received chemotherapy.
