WCLC 2026 | Biallelic inactivation of KEAP1/STK11 predicts response to checkpoint blockade in NSCLC

Mark Jeng, MD, Memorial Sloan Kettering Cancer Center, New York, NY, shares insights into a study evaluating the role of gene zygosity of KEAP1/STK11 in response to checkpoint inhibition in patients with non-small cell lung cancer (NSCLC). KEAP1/STK11 biallelic inactivation was associated with better response to dual checkpoint blockade, in particular in patients with KEAP1 biallelic inactivation. In patients with monoallelic inactivation, no difference was seen between response to single versus dual checkpoint blockade, indicating that zygosity plays a role in response and should be a consideration when deciding whether to treatment patients with dual immune checkpoint blockade. This interview took place at the IASLC 2026 World Conference on Lung Cancer congress in Seoul, Republic of Korea.
