ESMO GI 2026 | GLEAM: zolbetuximab plus gemcitabine and nab-paclitaxel in CLDN18.2-positive mPAC
Wungki Park, MD, MS, Memorial Sloan Kettering Cancer Center, New York, NY, discusses the background and findings from the Phase II GLEAM study (NCT03816163) of zolbetuximab plus gemcitabine and nab-paclitaxel in patients with CLDN18.2-positive metastatic pancreatic adenocarcinoma (mPAC). The trial did not meet primary endpoints but did find a higher objective response rate and duration of response in the zolbetuximab arm. In addition, treatment with zolbetuximab plus gemcitabine and nab-paclitaxel in patients who had elevated circulating interferon gamma-associated interleukin-18 was associated with a longer overall survival, highlighting a potential subgroup that zolbetuximab could be investigated further in. This interview took place at the European Society for Medical Oncology (ESMO) Gastrointestinal Cancer 2026 Congress in Munich, Germany.
