Avantika Gupta, PhD, Memorial Sloan Kettering Cancer Center, New York, NY, comments on the role of APOBEC3 mutational signatures as biomarkers for poor treatment outcomes in metastatic breast cancer. In preclinical models, APOBEC3 mutagenesis promotes resistance to endocrine and targeted therapies, including CDK4/6 and HER2 inhibitors. Genomic instability caused by APOBEC3 activity contributes to therapy-resistant mutations, such as RB1 loss. This interview took place at the 2024 San Antonio Breast Cancer Symposium (SABCS) congress in San Antonio, TX.
