SABCS 2024 | Overcoming resistance to trastuzumab deruxtecan in HER2-low cancers

Avantika Gupta, PhD, Memorial Sloan Kettering Cancer Center, New York, NY, examines mechanisms of resistance to trastuzumab deruxtecan (T-DXd), where efficacy is reduced despite strong results in HER2 -low breast cancer. Findings indicate EGFR overexpression limits T-DXd cellular uptake by restricting HER2 dynamics. Preclinical models demonstrate that combining T-DXd with EGFR monoclonal antibodies, such as cetuximab, enhances drug delivery and restores efficacy. This interview took place at the 2024 San Antonio Breast Cancer Symposium (SABCS) congress in San Antonio, TX.
