ESMO 2026 | Assessing the pan-RAS activity of GFH276 in various solid tumors

Yuxiang Ma, MD, PhD, Sun Yat-sen University Cancer Center, Guangzhou, China, comments on the preliminary results of a Phase I escalation trial (NCT07198321) of GFH276, which included various tumor types such as pancreatic cancer and non-small cell lung cancer with a limited number of patients and different dose ranges. While pre-clinical data have shown promising anti-tumor activities in KRAS G12C, G12D, and G12V models, it is too early to draw conclusions on the activity in different subtypes due to the small number of patients in certain subtypes. This interview took place at the European Society for Medical Oncology (ESMO) 2026 Congress in Madrid, Spain.

What I report today is the part one escalation. Escalation including a lot of tumor types, including pancreatic cancer, non-small cell lung cancer, and colorectal cancer. And the number is limited, and with different dose ranges, from 50 milligrams to 320 milligrams. So when you do the subgroup analysis for those subtype patients, it’s really a very small number of patients in certain subtypes for certain cancer types malignancy. So I couldn’t draw any conclusion of the activities in different subtypes. We can see the pre-clinical data released earlier last year in AACR 2025 and AACR 2026. It shows very promising anti-tumor activities in pre-clinical models in KRAS G12C, G12D, and G12V. But if we do a subgroup analysis in this trial for the escalation, it’s quite easy, quite early to jump to a conclusion. Maybe we can wait until future when we have more data in phase 1B expansion. We can have some clues.
