ESMO GI 2026 | Can ctDNA guide cetuximab sequencing and re-challenge strategies in MSS mCRC?

Giulia Martini, MD, PhD, Università degli Studi della Campania Luigi Vanvitelli, Naples, Italy, comments on results from the CAPRI-2 GOIM trial (NCT05312398) evaluating plasma circulating tumor DNA (ctDNA) comprehensive genomic profiling to guide sequential cetuximab-based therapy in RAS/BRAF wild-type microsatellite stable (MSS) metastatic colorectal cancer (mCRC). Genomic profiling at baseline and following first-line treatment allowed for selection of patients that could really respond to EGFR-targeted treatment. In the trial, later lines of treatment could be selected, so hyperselection could help clinicians select patients for re-challenge strategies. This interview took place at the European Society for Medical Oncology (ESMO) Gastrointestinal Cancer 2026 Congress in Munich, Germany.
