Yelena Janjigian, MD, Memorial Sloan Kettering Cancer Center, New York, NY, discusses the 5-year follow-up results from the CheckMate 649 trial (NCT02872116) of first-line nivolumab plus chemotherapy versus chemotherapy alone for advanced non-HER2+ gastric, gastroesophageal junction, or esophageal adenocarcinoma. Nivolumab-based therapy sustained improvements in overall and progression-free survival across all PD-L1 subgroups and randomized patients. The regimen also demonstrated higher and more durable response rates without new safety concerns. These findings reinforce nivolumab plus chemotherapy as a standard first-line option in this setting.
