SITC 2022 | Ultra-pH sensitive nanoparticles increasing therapeutic index of IL-2-Fc

Jinming Gao, PhD, UT Southwestern Medical Center, Dallas, TX, discusses the use of ONM-405, a hydrophobic micelle delivery system of IL-2-Fc, to drive an ultra-pH sensitive response for drug delivery to solid tumors. Micelle assembly and disassembly was induced within a quarter pH, less than 2-fold proton concentration, compared with conventional small molecule pH sensors and sensitive polymers which require 2 pH units, a 100-fold proton concentration, in order to get a ten-fold signal. This design increases pH sensitivity and allows an almost binary response to target specific pH thresholds. This interview took place at the 37th Annual Meeting of the Society for Immunotherapy in Cancer (SITC 2022) in Boston, MA.
