ASCO 2026 | Prior CTLA-4 blockade linked to immunosuppressive signature in melanoma
Omid Hamid, MD, The Angeles Clinic and Research Institute, Los Angeles, CA, discusses the impact of prior anti-CTLA-4 blockade on subsequent immunotherapy response in patients with anti-PD-1 refractory metastatic malignant melanoma enrolled in a Phase I/II trial (NCT04123470). Prior anti-CTLA-4 treatment within 12 months was associated with a hyperactive immunosuppressive signature and markedly worse survival outcomes, highlighting the need for further evaluation of washout periods and sequencing strategies following anti-CTLA-4 therapy.
