GU Cancers 2026 | Assessing darolutamide and ADT safety vs other ARPIs in mCSPC

Neal Shore, MD, FACS, Carolina Urologic Research Center, Myrtle Beach, SC, comments on a matching-adjusted indirect comparison evaluating the safety profile of darolutamide plus androgen deprivation therapy compared with enzalutamide, apalutamide, and abiraterone in metastatic castration-sensitive prostate cancer (mCSPC). The analysis incorporated individual patient data from ARANOTE (NCT04736199) alongside aggregate data from ARCHES (NCT02677896), TITAN (NCT02489318), and LATITUDE (NCT01715285) trials. Darolutamide demonstrated favorable safety outcomes with significantly lower rates of fatigue and rash versus comparators, supporting its differentiated tolerability profile in this treatment setting. This interview took place at the 2026 ASCO GU Cancers Symposium in San Francisco, CA.
