ELCC 2026 | How does disease biology vary across BRAF mutation classes in lung cancer?

Mariana Brandao, MD, PhD, Jules Bordet Institute, Brussels, Belgium, describes the three classes of BRAF mutations in lung cancer. Class I mutations are BRAF-activating mutations, forming RAS-independent monomers that are sensitive to first-generation inhibitors. Class II mutations form RAS-independent dimers and are insensitive to first-generation inhibitors, due to the RAF inhibitor paradox where these inhibitors activate BRAF. Class III mutations result in a low activity or inactive protein that binds to RAS more tightly, amplifying the downstream signal. This interview took place at the 2026 European Lung Cancer Congress in Copenhagen, Denmark.
