Susanna Ulahannan, MD, University of Oklahoma Health Sciences Center, Oklahoma City, OK, discussed first-in-human findings from the ART0380-01 trial (NCT04657068) of the ATR inhibitor ART0380 with low-dose irinotecan in advanced solid tumors. The regimen was well tolerated and showed encouraging antitumor activity, particularly in patients with ATM-deficient tumors. The dual strategy of exploiting replication stress and ATR inhibition produced objective responses and manageable toxicity. These results support further investigation using biomarker-driven selection to refine efficacy in this molecularly defined population.
