ASCO GI 2025 | Characterizing MTAP-deleted tumors in GI cancers using the Tempus database

Jun Gong, MD, Cedars-Sinai Medical Center, Los Angeles, CA, reflects on the findings of a multi-institutional collaboration with Tempus aimed at characterizing MTAP-deleted tumors across gastrointestinal cancers to guide the development of PRMT5 inhibitors. MTAP deletions were most frequently identified in pancreatic cancer, followed by cholangiocarcinoma (CCA), gastroesophageal cancers, and lastly, colorectal cancer. Potential co-occurrence of MTAP deletions with BRAF alterations and FGFR2 fusions in CCA, as well as SMAD4 alterations across all GI cancers were also observed. Further immune profiling revealed a reduced presence of B cells and CD4 T cells, as well as additional CD8 T cell reduction in certain cancers. This interview took place at the ASCO GI Cancers Symposium 2025 in San Francisco, CA.
